Mankiewicz wurde 1909 als Sohn polnisch-amerikanischer Juden in Pennsylvania geboren, zog aber bald mit seiner Familie nach New York. Bevor er sich dem Film zuwendete, arbeitete er einige Zeit in Berlin als Auslandskorrespondent für die Zeitung "Chicago Tribune".
